Both purinergic signaling through nucleotides such as ATP and noradrenergic signaling through molecules such as norepinephrine regulate vascular tone and blood pressure. Pannexin1 (Panx1), which forms large-pore, ATP-releasing channels, is present in vascular smooth muscle cells in peripheral blood vessels and participates in noradrenergic responses. Using pharmacological approaches and mice conditionally lacking Panx1 in smooth muscle cells, we found that Panx1 contributed to vasoconstriction mediated by the α1 adrenoreceptor (α1AR), whereas vasoconstriction in response to serotonin or endothelin-1 was independent of Panx1. Analysis of the Panx1-deficient mice showed that Panx1 contributed to blood pressure regulation especially during the night cycle when sympathetic nervous activity is highest. Using mimetic peptides and site-directed mutagenesis, we identified a specific amino acid sequence in the Panx1 intracellular loop that is essential for activation by α1AR signaling. Collectively, these data describe a specific link between noradrenergic and purinergic signaling in blood pressure homeostasis.

The local application of FK506 via the local delivery system disclosed herein results in excellent nerve regeneration while preventing the toxicity of systemic FK506 that has prevented clinicians from using FK506 routinely for treating severe cases of peripheral nerve injuries. A stent provided at the proximal end of the stent graft includes loops of material that co-operate with restraining wires that extend between a central guide wire carrier and a restraining wire cannula. The constraining mechanism acts to maintain the proximal stent constrained at both the proximal and distal ends of the proximal stent. The proximal stent is thus allowed to expand after expansion of the remainder of the stent graft during deployment. In an embodiment, the constraining mechanism acts to constrain two adjacent struts of the proximal stent at three points radially therearound, at the proximal end of the stent and at the distal end of the stent. The proximal stent may then overlap with the interior of an adjacent stent at an inner part of a curved vessel.

The spacer tip or standoff sleeve further prevents collapse of the bubble and ingress of liquid into the laser path. The spacer tip or standoff sleeve may be a generally-cylindrical protective cap that is fitted to an end of the optical fiber and that extends beyond the fiber tip to provide a predetermined spacing or standoff between the fiber tip and the stone when the protective cap is in contact with the stone. Medical radiography requires specialist control of radiography equipment to achieve good imaging results. Typical errors that can occur consist of an inappropriate field of view being accidentally applied. This results in a “cropping effect” in which portions of the region of interest of a patient which would be of clinical use are omitted from the image. Conventionally, the only solution is to re-take the entire image with a more appropriate (and inevitably larger) field of view selected. This is undesirable, because it might require recall of the patient from another location, and the patient will be subject to two exposures, thus undesirably increasing their X-ray dosage. The present application proposes to use an anatomical atlas to analyse an X-ray image output from an initial exposure, in particular to assess whether significant anatomical elements are missing from the image. If elements are missing, the field of view of the X-ray imager is recalibrated, using collimation or pan/tilt adjustments, for example. A subsequent X-ray image is obtained, and combined with the initial image, to provide an output image. Because only a small area of the region of interest may subsequently need to be exposed, a smaller additional dose results.
